1,6-二磷酸果醣对兔心肌细胞中LDH漏出和ATP含量的影响
Effects of Fructose-1,6-Diphosphate on Rabbit Cardiomyocytes
【摘要】 为探讨 1,6 -二磷酸果醣 (FDP)对心肌细胞的影响 ,采用胶原酶二步灌流法将成年兔心肌细胞分离出来 ,培养于含有 2 5和 5 0mmol/LFDP的培养基中 ,分别于 0 ,4 ,8,12h取样 ,作台盼兰染色和光学显微镜上的观察 ,并对培养基中乳酸脱氢酶 (LDH)活性及心肌细胞中三磷酸腺苷 (ATP)含量进行了测定 .结果 ,在上述条件下 ,圆形细胞 (死亡的或受损后尚存活 ,但形态变圆的细胞 )的百分率一直保持恒定 .在FDP实验组 ,各时间点的LDH活性均低于对照组 .在 2 5mmol/L ,4h时 ,LDH的活性显著降低 (P <0 0 5 ) .而在 5 0mmol/L浓度 ,12h时 ,LDH活性却显著升高 (P <0 0 5 ) .在FDP低浓度为 2 5mmol/L和高浓度 5 0mmol/L时 ,与对照组相比 ,在各时间点 ,心肌细胞中ATP含量呈增加趋势 ,并在 5 0mmol/L浓度 ,8h时 ,差别显著 (P <0 0 5 ) .本研究表明 :在浸泡式培养的条件下 ,不同浓度的FDP ,在不同的接触时间 ,对兔心肌细胞膜产生不同的影响 .低浓度时 ,FDP对兔心肌细胞膜有稳定作用 ,浓度高 ,接触时间长 ,可造成心肌细胞膜通透性增高 ,进而造成心肌细胞损害 .结论 :无论将FDP用于临床医疗或科学研究中 ,根据用药目的 ,给药途径 ,方法及接触时间的不同 ,FDP的剂量可有很大差异 .因此 ,为满足使用需要 ,寻找出FDP的最适
【Abstract】 To investigate effects of fructose-1,6-diphosphate (FDP) on cardiomyocytes, rabbit cardiomyocytes were isolated by two steps collagenase perfusion method. The cells were cultured in the medium with 2 5 and 5 0?mmol/L FDP and assay of adenosine triphosphate (ATP) quantitation at 0,4,8,12?hours respectively. Samples were taken for trypan blue up take, round cell percentage calculation, test of lactate dehydrogenase (LDH) activity. The results from this study showed that the percentages of round cells were stable under the conditions of this study. The LDH activities in the FDP groups were lower than control groups, at different hours. Especially, the difference is significant in 2 5?mmol/L FDP group at 4?hours compared with control group(P<0 05). But the LSH activities in the 5 0?mmol/L FDP group at 12?hours was significant high (P<0 05). The ATP quantities in FDP groups at different hours showed the trend of increase. The ATP increase in 5 0?mmol/L FDP group at 8?hours was significant (P<0 05). The study indicated that different doses of FDP and different exposure durations caused different effects on soaked culture rabbit cardiomyocytes. Lower concentration of FDP kept cell membrane in stable condition. Higher concentration of FDP and longer exposure duration resulted in high permiability of cell membrane which can damage cardiomyocytes. It is suggested that the doses are quite big different according to the administrative way, the exposed duration and the purpose of usage when FDP is applied in clinic treatment and research work. So, it is important to find proper doses and exposure durations to meet different needs of usage.
