【摘要】 目的 :探讨中药葛根的新型制剂葛根素注射液抑制 (延缓 )自发性高血压大鼠 (SHR)肾脏小血管重塑的作用及其机制。方法 :选取 16周龄的自发性高血压大鼠为动物模型 ,以葛根素注射液治疗并与苯那普利对照。结果 :实验结束时 ,病理对照组血压较实验开始时显著上升 (P <0 .0 5 ) ,葛根素注射液无论对SHR的收缩压还是舒张压均有明显的降低作用 (P <0 .0 1) ,同时降低血浆内皮素 ,血清胰岛素 (P <0 .0 1,P <0 .0 5 ) ,与苯那普利比较无显著性差异 (P >0 .0 5 ) ,降低血浆血管紧张素Ⅱ优于苯那普利。光镜检查显示 ,病理组大鼠肾小动脉壁显著增生肥厚 ,管腔狭窄 ,内层弹力纤维松驰、断裂 ,而葛根素及苯那普利组大鼠肾小动脉管壁增生明显减轻 ,内层弹力纤维结构完整、致密 ,无松驰、断裂现象。结论 :葛根素注射液具有明显抑制 (延缓 )肾血管重塑的作用 ,该作用可能与降压 ,降低血浆血管紧张素Ⅱ ,内皮素水平及改善胰岛素抵抗有关。更多还原

【Abstract】 Objective:To study the effect of puerarin injection on restraining(or delaying)the reconstruction of the renal blood vessels in spontaneous hypertension rats(SHR)and explore its mechanism.Methods:Twenty-eight SHRs(sixteen-week-old)were randomly divided into three groups.The puerarin treating group(ten SHRs)were given abdominal injection (100 mg·kg -1·d -1)and the control groups were respectively given with benazepril (ten SHRs, 5 mg·kg -1·d -1)and water(eight SHRs)for six weeks.The blood pressure of tail artery was measured before the experiment,the second forth and sixth weeks.By the end of the experiment,the blood were gathered of serum Endothelin(ET),Angoitension-Ⅱ(AT-Ⅱ)and Insulin (Ins).The renal tissues were examined by light microscope to comprehend the pathobiologic changes.Results:By the end of the experiment the blood pressure of the pathology control group was significantly higher than that of the beginning(P<0.05).Puerarin injection can decrease not only the systolic blood pressure but also the diastolic pressure (P<0.05).The serum AT-Ⅱ,ET,Ins of puerarin treating group significantly decreased compared to the pathology control group(P<0.05).In the sight of the data,the puerarin seemed to have superiority in reducing the Ins.Severe hypertrophy and sclerosis on the walls of arterioles,narrow in the lumina,loose and break in inner elastic fiber were seen in the pathology control group,while the puerarin and benazepril could attenuate the above injuries of the renal small arteries.Conclusion:Puerarin injection can significantly restrain (or delay)the reconstruction of the renal blood vessels.Reducing the blood pressure and the level of the serum AT-Ⅱ,ET and improving the Ins resistance may be involved in the mechanism concerned.更多还原
