热休克蛋白的产生提高大鼠离体皮瓣的成活
Induction of heat shock protein improved free museulocutaneous flap survival in rats
【摘要】 观察热休克蛋白对离体皮瓣成活的影响。雄性大鼠分对照组 ,全身加热组 ,局部加热组。加热组分别全身或局部 4 2℃作用 30min ,离体皮瓣取自大腿内收缩肌条一块 (2cm× 6cm) ,植入同侧腹股沟。手术 3d后处死动物 ,用硝基蓝四氮唑染色判定皮肤损失面积 (cm2 )及肌肉运动性 ,皮瓣上皮肤和肌肉用定量Norther和Wester分析HSP72 的变化及离体皮瓣存活率。局部和全身预热的大鼠皮肤成活率明显提高 2 3% ,HSP72 的表达升高 ,与对照组相比P <0 0 5。实验结果表明热诱导热休克蛋白的产生有利于皮肤移植后皮瓣的成活。
【Abstract】 To determine the heat shock protein response in skin and muscle after systemic or local heating and to examine the effect on free musculocutaneous flap survival in a rat model.Free musculocutaneous flaps incorporating thigh adductor muscles and a 2 cm×6 cm skin paddle were transplanted into ipsilateral groin in three groups of male wistar rats. Systemically heated rats and Locally heated rats were anesthetized and incubated for 30 minutes at 42℃ 6 hours before free musculocutaneos tissue transfer. Control rats did not have heating pretreatment but received identical anesthesia.Animals were sacrificed on postoperative day 3, and then the skin loss(cm 2) and muscle viability were quantified by nitroblue tetrazoliun staing time in a blinded fashion. The skin and muscle from the free flap were analyzed for HSP 72 mRNA and protein content with quantitative Northern and western blot techniques. All free musculocntaneous flaps were viable. However,the locally and systemically heated rats demonstrated a marked improvement of skin survival,which was correlated with increased skin levels of HSP 72 . There were differences in nitroblue tetrazolium muscle staining times or muscle levels of HSP 72 among the three groups. These results demonstrated that prior heat induced heat shock proteins improved in musculocutaneous flap survival.
