【摘要】 探讨银杏叶提取物 (GBE)对脂多糖 (LPS)诱导大鼠急性肺损伤 (ALI)的保护作用。给大鼠静注LPS复制ALI动物模型。首先动态观察ALI的形成过程 ,注LPS后 2h已形成明显的ALI。另批大鼠随机分成 :对照组 (静注生理盐水 ) ,LPS组 (静注LPS ,5mg kg体重 ) ,GBE +LPS组 (注LPS前三天开始每天灌胃给GBE一次 ,按黄酮甙 8mg kg体重计算 ,当日在给LPS前 2h再给一次GBE) ,每组 6只大鼠。注LPS或盐水后 2h收集标本 ,进行测定。发现 :注LPS后肺湿 干重量比 ,肺泡灌洗液中蛋白含量及肺血管通透指数均升高 (均P <0 0 1) ;血中乳酸 (LD) ,一氧化氮(NO) ,内皮素 1(ET 1) ,肿瘤坏死因子 α (TNF α)含量及血乳酸脱氢酶 (LDH)活性 ,均有显著升高 (P <0 0 1,P <0 0 0 1)。而肺组织细胞膜Na+ K+ ATPase活性显著下降 (P <0 0 1)。预先给予GBE可显著缓解上述变化 (P <0 0 5 ,P <0 0 1)。提示GBE在防治肺的急性炎症性疾病中有一定应用前景。更多还原

【Abstract】 To investigate the protective effect of Gingko Biloba extract (GBE) on LPS-induced acute lung injury(ALI) in rats. ALI animal model was reproduced by intravenous injection LPS( 5mg/kg BW). The dynamic process with the time after injection LPS was observed. Obvious ALI was developed after 2 hours of injection. Another 18 rats were randomly divided into three groups. 1.control rats ( saline), 2. LPS rats (LPS,5mg/kg BW), 3. GBE+LPS rats (GBE was given 3 days before LPS, once a day via the esophagus, according to the amount of flavone glycosides 8mg/kg BW ).There were 6 rats in each group. The samples were collected after 2 hours of LPS or saline administration. Results showed that: In LPS rats, ratio of wet/dry lung weight, protein content in BALF and pulmonary permeability index were obviously increased(P<0.001).LD, NO, ET-1? TNF-α content in blood all increased significantly (P<0.01 or P<0.001). But, Na +-K +-ATPase activity in cell membrane of lung tissue was obviously decreased(P<0.01).The changes of above parameters were obviously attenuated by GBE (P<0.05,P<0.01). Results suggested that GBE has certain protective effects on ALI in rats and may be useful in the treatment of acute pulmonary inflammatory disease.更多还原
