【摘要】 目的　从分子水平检测中药预防肝纤维化的作用及机制。方法　利用 CCl4 复合因素对SD大鼠复制肝纤维化模型 ,用中药灌胃预防 ,检测其生化指标、肝纤维化指标、病理情况以及肝组织中 HGF m RNA表达量。结果　肝纤维化组及中药预防组的 AL T、AST、A/ G均较空白组恶化 (P <0 .0 1) ,但中药预防组的 AL T、AST、A/ G的恶化程度低于肝纤维化组 (P <0 .0 1)。 HA(透明质酸 )、PC ( 型血清前胶原 )、L N (层粘蛋白 )、PL D(羟脯氨酸 )四项血清学指标的结果显示 :肝纤维化组及中药预防组均高于空白组 (P <0 .0 1) ,但中药预防组的升高幅度低于肝纤维化组 (P<0 .0 1) ,此四项指标的降低与病理组织切片反应肝纤维化的程度相一致。中药预防组与肝纤维化组肝组织 HGFm RNA的相对表达量比空白组高 (P <0 .0 1) ,中药预防组肝组织 HGFm RNA的相对表达量较肝纤维化组高 (P <0 .0 1)。结论　联合检测生化、肝纤维化指标可提高肝纤维化早期诊断 ;肝纤维化时肝组织 HGF m RNA的表达增加 ,中药通过促进肝组织 HGF m RNA的表达可延缓或减轻肝硬化的发生更多还原

【Abstract】 Objective To study the mechanism of preventive effect of Chinese herbs on the CCl 4 induced hepatic fibrosis at molecular level. Methods After hepatic fibrosis model of rat was established with CCl 4 and Chinese herbs were administrated at the same time of CCl 4 injection, the level of HGF mRNA expression,biochemical and hepatic fibrosis parameters, and pathologic changes were determined. Results The concentration of ALT,AST, HA, LN, PLD and PCⅢ in serum of hepatic fibrosis rat, and the level of HGF mRNA expression in liver of hepatic fibrosis rat determined by RT PCR increased compared with those of control group, while A/G decreased(P<0.01). After Chinese herbs were administrated by mouth into rat at the same time of CCl 4 injection, the level of HGF mRNA expression in liver cells was higher, but the concentration of HA, LN, PLD and PCⅢ in serum were lower than those of the group of hepatic fibrosis model, while A/G was higer(P<0.01). Conclusions Connecting the biochemisty and hepatic fibrosis parameters, the early diagnosis could be made. Increasing of HGF mRNA in rat liver was involved when hepatic fibrosis occurred, and Chinese herbs could prevent hepatic fibrosis by promoting HGF gene expression.更多还原
