石墨电极固载DNA的一种新修饰方法
A New Method for Surface Modification of Paraffin Impregnated Graphite Electro de for DNA Immobilization and Hybridization
【摘要】 首次报道通过充石蜡石墨电极电化学氧化腐蚀制成的微孔穴电极 ,成功地用于ss DNA的固载和杂化反应 ,证明了这种微孔穴石墨充石蜡电极可以固载较多量的ss DNA(包括解链的小牛胸腺DNA和 15 merss DNA) ,可以成为性能良好的DNA杂化探针 .用Co(phen) 3 + 3 电化学指示剂和循环伏安方法 ,讨论了电极预氧化腐蚀的时间和ss DNA固载时间的影响 ,提出了DNA探针在微孔穴三维结构中固载的可能机理
【Abstract】 A micro-hollow paraffin impregnated graphite elec tr ode (MHPIGE) was developed by electro-oxidation of a paraffin impregnated graph ite electrode for immobilization of DNA. Immobilization of single string DNA fro m CT-DNA and 15 mer ss-DNA onto the prepared MHPIGE was investigated by cyclic voltammetry using Co(phen)~3+3 as an indicator. It was demonstrated that the MHPIGE could immobilize significant amounts of ss-DNA, but not ds-DNA, un der mild conditions. The immobilized ss-DNA underwent hybridization reactio n very well. The effects of duration on pre-oxidation treatment and on ss-DNA immobilization reaction were investigated. The mechanism of DNA immobilization o n the MHPIGE is proposed as an effect of the specifically formed three-dimensio nal micro-hollow structure.
