利用酶反应热效应检测过氧化氢酶活性
Detecting Catalase Activities with Exothermic Reaction
【摘要】 为了探索酶活性测定的新方法 ,降低酶活性常规测试中干扰因素的影响 ,提高酶活性测定的准确性 ,利用低电势电位差计及热电偶换能器检测过氧化氢酶活性 .结果表明 ,测定时的 H2 O2 质量分数以 9.6 %为好 ,可以用于酶活性从 0 .5 u至 3.5 u酶活力的检测 ;当 H2 O2 质量分数在 2 .4 %~ 7.2 %时 ,酶反应速率与底物浓度有线性关系 ;当底物质量分数超过 9.6 %时 ,酶反应速率显现底物抑制的动力学特征
【Abstract】 To explore a new method of determing enzyme activities,and reduce the disturbing factors’ effects on this determination,and raise the accuracy of enzyme determination,low electropotential meter and thermocouple are used to determine catalase activities. While the concentration of H 2O 2 is 9.6%,the enzyme activities from 0.5 u to 3.5 u can be detected;while H 2O 2 concentration from 2.4% to 7.2% ,the enzyme reaction velocity is in direct proportion to substrate concentration. Above 9.6% substrate concentration,the velocity of the enzyme reaction shows the kinatic character of the substrate inhibition.
