甲状腺肿块中端粒酶活性检测及其意义

【摘要】 目的 研究甲状腺肿块 (甲状腺癌及甲状腺腺瘤 )中端粒酶活性与甲状腺肿块性质的关系。方法 采用PCR新的高灵敏度的方法 (TRAP法 ) ,检测 4 0例甲状腺肿块标本 (2 5例甲状腺癌 ,15例甲状腺腺瘤 )端粒酶活性 ,同时以L1、A1肺癌细胞株和正常的甲状腺组织作为活性阳性和阴性的对照组。结果 在 2 5例甲状腺癌标本中有 2 1例发生了阳性反应 (占 84 % ) ,而 15例甲状腺腺瘤标本中仅 3例发生了阳性反应 (占 2 0 % ) ,差异十分显著 (P <0 0 1)。 2 1例甲状腺癌阳性反应病例中 ,端粒酶活性反应与肿瘤的类型、病程、病情等有关 ;3例甲状腺腺瘤阳性反应病例中 ,端粒酶活性反应与腺瘤组织结构分化异常有关。结论 本研究结果显示 ,端粒酶常发生于甲状腺恶性肿瘤组织中 ,并在甲状腺组织恶性变作用中起着重要作用。在甲状腺组织微量检测到 ,端粒酶活性检测有望成为一种良好的诊断方法
【Abstract】 Objective To investigate the presence of telomerase activity in thyroid tumors (cancer, adenoma) and the relationship between telomerase activity and human thyroid tumors. Methods 40 human thyroid tumor samples (25 thyroid cancer and 15 thyroid adenoma) were assayed for telomerase activity using a highly sensitive PCR technique (TRAP). At the same time, L 1, A 1 lung cancer cell samples and normal thyroid tissue samples were used as the positive and negative control groups of telomerase activity respectively. Results Telomerase activity was detectable in 21 of 25 thyroid cancer (84%), but in 3 of 15 thyroid adenoma (20%). There was a significant difference between the two group using the standard u test of rate (P<0.01). The telomerase activity intensity in 21 positive cancer samples was interrelated with the character of tumor highly, such as the type, degree, size and differentiation of the tumor, etc. The expression of telomerase activity in 3 positive adenoma samples was associated with phenotypic progressing and degree of dysplasia. Conclusion Our study indicated that telomerase was activated frequently in thyroid cancer and might play an important role in thyroid carcinogenesis. The telomerase activity might be a potential diagnosis assay for human thyroid tumor especially for microtissue of thyroidea.
