背根神经节慢性压迫大鼠行为及缰核内c-fos CGRP的变化
Changes of Behavior and CGRP, c-fos in Habenula after Chronic Compression of DRG in Rats
【摘要】 目的 建立有效的背根神经节压迫模型 ,并探讨缰核内c -fos和CGRP的变化。方法 采用铬制 1- 0肠线通过椎间孔压迫Wistar大鼠L4 、L5背根神经节制备背根神经节压迫模型 ;应用SP法检测缰核c -fos表达 ;放射免疫分析法测定缰核内CGRP含量。结果 术后 4周内受压侧后肢持续表现痛敏行为 ,制备了稳定的背根神经节压迫模型 ;外侧缰核检测到中等强度表达的核内fos蛋白免疫阳性细胞 ,其表达呈一过性 ;测得缰核内CGRP含量基础值为 (5 0 6 6± 0 4 34)ng/L ,并随压迫刺激时间的延长 ,CGRP呈逐渐增高趋势。结论 本实验建立了慢性背根神经节压迫的可靠模型。缰核内c -fos表达呈一过性增高 ,而CGRP呈持续增高 ,与痛敏行为呈负相关 ,说明缰核参与慢性痛的形成与调制
【Abstract】 Objective To develop a reliable and reproducible model of hyperalgesia arising from the dorsal root ganglion and to understand the function of habenula in chronic pain. Methods 1-0 chronic gut sutures were inserted into the left L 4, L 5 intervertebral foramams to establish the model; c-fos expression was used th judge the reactivity of habenula neurons to the chronic pain; CGRP contents were quantified in habenula by radioimmunoassay (RIA). Results The model was successful when mechanical hyperalgesia occurred in rats of compression of DRG. 4 weeks after compression;c-fos expression was transiently increased in the lateral habenula; CGRP in habenula was significantly elevated and remained the tendency along the period of 4 weeks. Conclusion This experimental model reliably produced a disorder resembling clinical low back pain; c-fos expression was transiently increased while CGRP kept level in habenula during chronic low back pain that suggests habenula might be an important relay of chronic pain.
