农杆菌介导Barnase基因转化番茄

【摘要】 以雄性不育基因barnase作为目的基因、抗除草剂溴苯腈的bxn基因作为选择标记基因 ,利用下胚轴和子叶做外植体与农杆菌共培养方法 ,在番茄的两个品种佳粉 1号和佳粉 15号中 ,分别获得 38和 15株溴苯腈的抗性株 .PCR检测结果表明 ,抗性株含barnase基因 ,且抗性株的花粉完全丧失活力 ,证实所得的溴苯腈抗性株为转Barnase基因番茄

【Abstract】 Using transformation technique, some male sterile gene transformants were obtained in tomato (Lycopersicon esculentum Mill.). The Agrobacterium tumefaciens strain used was LBA4404 containing a male sterile gene barnase and a selectable marker gene bxn which confers resistance to the herbicide bromaxynil. Tomato male sterile transformants were acquired via tissue culture in vitro using the cotyledonary and hypocotyl segments as the explants. 38 and 15 transformants were obtained in two varieties, Jiafen 1 and Jiafen 15 respectively. They regenerated and rooted on the medium with 0.407 mM bromaxynil. PCR results are positive with perfect resistance to the bromaxynil.
