果蝇基因组DNA的大规模提取及其RAPD-PCR反应条件的优化
Large-scale extraction of genomic DNA from Drosophila and optimization of its RAPD-PCR
【摘要】 介绍了一种大规模提取高质量果蝇基因组DNA的方法,该法不需液氮研磨,不需匀浆转移,不需高速离心,全部操作在3个1.5μL离心管中完成,所以特别适用于对不同果蝇品系等多样本的大规模提取.以本法提取的DNA为模板建立了RAPD-PCR分析的最佳反应体系,获得了稳定而理想的扩增效果,特别适用于小型昆虫的群体遗传多样性研究.
【Abstract】 A method large-scale extraction of high quality genomic DNA from Drosophila is reported. This method need not grinding flies with liquid nitrogen, transferring the serosity and high speed centrifuging, all the operations are finished in three 1.5?μL micro-tubes .which is most applicable for large-scale DNA extraction from different samples. Using the DNA extracted by this method as template, established the optimum conditions for RAPD-PCR(random amplified polymophic DNA-polymorase chain reaction), which can be applied to genetic diversity studies of little insects.
