间尼索地平光解产物的分离和结构

【摘要】 目的分离、制备间尼索地平的光解产物 ,确定其结构。方法将光照分解的间尼索地平乙醇溶液用TLC法找出分离条件 ,用柱层析制备光解产物 ,经1HNMR谱确认其结构 ;用化学法合成其参照物 ,进一步证实其结构。结果光解产物为 2 ,6 二甲基 4 (4 硝基苯基 ) 3,5 吡啶二羧酸甲酯异丁酯。结论间尼索地平乙醇溶液对光不稳定 ,生产过程应避免光照 ,在光解过程中二氢吡啶环被氧化成吡啶环的同时 4 位苯环上的硝基发生重排。

【Abstract】 ObjectiveTo separate a photodegradation product and to study its structure.MethodsThe solution of m-nisoldipine in ethanol exposed to sunlight for 12h, the favorable separative condition was studied via TLC. The solution irradiated was chromatographed on silica column with chloroform:ether (85:15) as eluant. The solutions obtained from the column were concentrated under reduced pressure to give an oily substance that was identified by NMR spectroscopy.ResultsSunlight irradiation of m-nisoldipine caused oxidation of the dihydropyridine ring and transmutation of the nitro group in nitrobenzene ring.ConclusionThe solution of m-nisoldipihe is not stable under lighting conditions. The regularity of rearrangement will be studied.
