钝顶螺旋藻DNA中G,C的质量分数的测定
Study on the Percentage of G.C in DNA of Spirulina Platensis
【摘要】 用Zarrouk培养基培养钝顶螺旋藻NS - 90 0 2 0 ,从中提取DNA ,然后在加热条件下 ,使其DNA变性 ,跟踪DNA的变性过程中的增色效应 ,得到螺旋藻的熔解温度 (Tm) ,根据经验公式 (G +C) % =2 .4 4 (Tm - 6 9.3)可以算出螺旋藻体内G ,C百分质量分数 ,作为进一步研究螺旋藻的遗传特性及育种的基础
【Abstract】 Spirulina platensis NS -90020 was cultivated in Zarrouk medium and its DNA was extracted and purified. Each absorption value was measured when the purified solution was heated at different temperatures. Then the percentage of G,C contained in DNA of spirulina platensis was calculated based on the equation of(G+C)%=2.44* (Tm-69.3) .This result can be used for further research on molecular biological characteristics of spirulina platensis.
