尖锐湿疣挖空细胞的原位杂交与电镜观察
A Study on the Koilocytes of Condyloma Acuminatum by Electronic Microscopy in Situ Hybridization
【摘要】 目的 :探讨尖锐湿疣挖空细胞的性质和形态变化 ,为病理诊断提供信息。方法 :采用东山区人民医院尖锐湿疣活检 5 0例。每例分别取材作透射电镜、原位杂交组化和HE染色光镜观察 ,将电镜所见的结果与原位杂交组化所得信息进行对比分析。结果 :发现所有挖空细胞核内均有HPV DNA存在 ,电镜下核内均含有多少不等染色质间颗粒 ,其中 3例含有病毒颗粒。核皱缩 ,染色质边集 ,深染 ,核仁消失。在HE切片上 ,挖空细胞单个、灶状或成片分布 ,核呈星形或毛虫样的形态特征。结论 :挖空细胞具有HPV感染引起表皮细胞凋亡形态特征 ,据此特征即可确定尖锐湿疣的病理诊断。
【Abstract】 Objective:In approaching the nature and morphogenesis of the koilocytes in condyloma acuminatum to provide understanding for pathologic diagnosis. Method:The biopsy of 50 cases of comdyloma acuminatum in Dongshan District Peoples Hospital were studied through ultrasecton for transmitting electronic microscopy,paraffin embed section for HE stain and in situ hybridization stains in every case. The changes of koilocytes under electronic microscopy were compared with findings of in situ hybridization.Result:The nuclei of all koilocytes were found to contain HPV DNA by in situ Hybridization. Interchromatin granules or virus granules,nucleous contract,chromatin aggregation under nucleous membrane,chromatism and nucleoli disapperance under electronic microscopy. On HE section koilocytes reaveled the special morphology of apoptosis,located in the epithelium singly or ingroups,and the unclei became star like or caterpiller like.Conclusion:The koilocytes possess the morphological speciality of epithelial apoptosis induced by HPV,according to which,the diagnosis of condyloma acuminatum can be determined.
