固态发酵生产腺苷酸脱氨酶

【摘要】 对多株曲霉产腺苷酸脱氨酶的性能进行了比较 ,发现米曲霉 3.80 0 (Aspergillusoryzae)产酶水平较高。该菌株固态发酵产酶的适宜培养基为 :以麸皮为主原料 ,蔗糖 2 % ,鱼粉 2 % ,(NH4 ) 2 SO4 0 .1% ,柠檬酸钠 0 .2 % ,MgSO4 0 .0 5 % ,吐温 - 80 0 .1% ,含水量 5 0 %。最佳的培养条件为 :2 5 0mL三角瓶装 2 0g培养基 ,在 2 8~ 30℃培养 6 0h。在优化条件下 ,培养物酶活可达到15 4 3.4 8u/ g鲜曲。

【Abstract】 Compared with several strains producing AMP deaminase, Aspergillus oryzae 3.800 was the best one. Its optimal composition of culture medium for enzyme secretion by solid state fermentation was: wheat bran as main material, sucrose 2%, fish meal 2%, (NH 4) 2SO 4 0.1%, Na 3C 6H 5O 7-2H 2O 0.05%, MgSO 4 0.05%, polysorbatum 80 0.1%, water hold 50%. The optimum cultivating conditions were as follows: 20g medium in 250mL flask, incubation temperature 28~30℃, the fermentation was stopped after 60h. In this way,the enzyme activity of moldy bran could reach 1543.18u/g.
