何首乌组培快速繁殖技术的研究
Study on Polygonum multiflorum rapid propagation technique
【摘要】 对何首乌的组培快繁技术进行了研究。结果表明 :采用 MS+6-BA1 .0 mg/L+IBA0 .1 mg/L的培养基能成功地诱导芽的分化。对于芽增殖 ,以 MS+6-BA 0 . 75 mg/L+IBA 0 .0 5 mg/L或MS+6-BA 1 .5 mg/L+IBA 0 .1 mg/L培养基较好 ,培养 3 0 d后芽增殖率可分别达到 3 .89和 4.1 1。诱导生根以 1 /2 MS+IBA 0 .5~ 1 .2 5 mg/L或 1 /2 MS+NAA 0 .5 mg/L培养基较好 ,培养 2 0 d后生根率可达80 .0 %~ 86.7%。
【Abstract】 In order to use the technique of tissue culture in the rapid propagation of Polygonum multiflorum and lay a foundation for the rapid propagation of its improved breeds,a series experiment had been done.The explants used for culture were stem segments with axillary bud of Polygonum multiflorum .The media were MS basal media containing different plant hormones.The results indicated that shoots could be induced successfully in MS medium supplemented with 6-BA1 0mg/L and IBA 0 1mg/L.The more suitable medium for bud multiplication was MS+6-BA 0 75 mg/L+IBA 0 05 mg/L or MS+6-BA 1 5 mg/L+IBA 0 1 mg/L.After 30 days’ cultivating,the propagation coefficient of the bud could reach to 3 89 and 4 11.The best culture medium for root inducement was 1/2MS+IBA 0 5~1 25 mg/L or 1/2MS+NAA 0 5 mg/L,and it could make 80 0%~86 7% culture young plants root in 20 days.
