硫酸铵盐析在蕲蛇毒凝血酶样酶分离中的应用
Application of ammonium sulfate salting-out to isolate thrombin-like enzymes from Agkistrodon acutus venom
【摘要】 探讨了硫酸铵沉降对蕲蛇毒中凝血酶样酶的初步分离效果的影响 ,确定出最佳处理条件 :2 .0 %的蕲蛇毒在pH 5 .0及 4 5 %饱和度的硫酸铵溶液中盐析沉降蕲蛇毒中非凝血酶样酶杂蛋白 ,上清液即为所分离的凝血酶样酶组分
【Abstract】 The method of ammonium sulfate salting-out to preliminarily isolate thrombin-like enzymes from Agkistrodon acutus venom was applied and the separation effect was discussed in this paper. The optimum treatment to salting-out non-thrombin-like-enzyme components in 2.0% crude venom was determined as 45% ammonium sulfate at pH?5.0. The supernatant is preliminary isolated thrombin-like enzymes. The specific activity of the isolated thrombin-like enzymes is three times as high as rude Agkistrodon acutus venom, and percent recovery of protein is 54.6%.
