分光光度法测定环氧化物浓度与环氧化物水解酶活性
A Spectrophotometric Assay for Determination of Epoxide Concentration and Epoxide Hydrolase Activity
【摘要】 研究了在弱碱性条件下 ,利用环氧化物与 4- (对硝基 )苄基吡啶之间的显色反应测定环氧化物含量 ,进而计算环氧化物水解酶活性的分光光度法。对于缩水甘油苯基醚 ,其最低检出浓度为 4.5 μmol/L,所生成染料的摩尔吸光系数 ε为 2 .2 0× 1 0 3L· mol- 1· cm- 1。在 4.5~ 30 0 μmol/L的浓度范围内 ,测量的精密度可达 0 .9%。
【Abstract】 A spectrophotometric method for determination of epoxide concentration and epoxide hydrolase activity has been developed by using the reaction of epoxide with 4 ( p nitrobenzyl) pyridine followed by alkalization. The lowest detectable concentration of slycide phenyl ether and the molar absorption coefficient of the dye were 4.5 μmol/L and 2.20×10 3 L·mol -1 ·cm -1 , respectively. The precision of the assay for phenyl glycidyl ether within the concentration range of 4.5~300 μmol/L was 0.9 %.
