一种基于适配分子的偏振荧光检测法
A Fluorescence Polarimetric Method for the Determination of Thrombin Based on Its Interaction with Aptamer
【摘要】 凝血酶适配分子 (aptamer)可与凝血酶以高亲和力结合 ,二者的结合可改变荧光素标记的适配分子溶液的荧光偏振强度 ,据此建立了适配分子荧光偏振法测定凝血酶的新方法。方法的线性范围是 0 .1~ 4IU/mL ;检出限为 0 .0 9IU/mL。将该方法用于人体血浆中凝血酶含量分析 ,结果满意
【Abstract】 The anti thrombin aptamer can interact with thrombin in high affinity. This interaction results in the change of the fluorescent polarization in the solution of the aptamer labeled with fluorescence. We developed a new method based on the change of the fluorescent polarization resulted from this interaction to detect thrombin. The linear range of the determination is 0.1~4 IU/mL. The detection limit for thrombin is 0.09 IU/mL. This method has been used to determine thrombin in human whole blood. The results obtained are satisfactory.
