骨髓基质细胞在三种可吸收生物膜上附着及增殖的比较

【摘要】 目的 比较骨髓基质细胞 (MSC)在三种可吸收生物膜上的附着及增殖情况 ,探讨三种可吸收生物膜作为组织工程支架材料的可行性。 方法 取 2月龄长枫雄性白猪 ,抽取髂骨骨髓 ,培养骨髓基质细胞 ,体外扩增后 ,取第 2代细胞种植于国产胶原膜、Bio Mesh R○ 及 Bio- Guide R○ 膜的表面 ,并利用 MTT方法检测种植后 1~ 8天细胞的活力。 结果  MSC种植后 1天 ,在三种膜上的附着差异无显著性 (P>0 .0 5 ) ,MSC在国产胶原膜及 Bio-Guide R○上生长、繁殖明显好于 Bio Mesh R○ (P<0 .0 5 )。 MSC在 Bio- Guide R○上第 2天开始细胞数量明显增多 (P<0 .0 5 ) ,一直增殖到第 5天趋于平稳 ;MSC在国产胶原膜上第 3天开始细胞数量明显增多 (P<0 .0 5 ) ,一直增殖到第 5天后趋于平稳 ;MSC在 Bio Mesh R○上生长到第 8天时 ,细胞数量仍无明显变化 (P>0 .0 5 )。 结论  Bio-Guide R○ 及国产胶原膜能够维持 MSC的生长与繁殖 ,在组织工程学领域 ,是一种有潜力的载体材料。

【Abstract】 Objective To compare the attachment and proliferation of marrow stromal cells(MSC) cultured on three kinds of bioabsorbable membrane in vitro in order to understand the biocompatibility of these membranes and their suitability for tissue engineering scaffold. Three commercially available collagen membrane, BioMesh R○ and Bio Guide R○ were examined. Methods These membranes were punched to form circular plates of 6 mm diameter and being put into tissue culture wells. MSC of the 2nd transferred culture were seeded into tissue culture wells with or without membrane. The rates of cell attachment and proliferation were measured with MTT colorimetric assay. Results No significant difference were observed between attachment levels for MSC cultured on three kinds of bioabsorbable membrane in long term attachment(24 h). The rates of cell proliferation differed among the membranes examined. Bio Guide R○ demonstrated a significantly higher rate of proliferation from 2 to 5 days. Collagen membrane demonstrated also a significantly higher rate of proliferation from 3~5 days. There was no significant proliferation of cells cultured on BioMesh R○ membrane. Conclusion Bio Guide R○ and Collagen membrane can support the growth and stimulate the proliferation of MSC in vitro and thus, are potential carriers for the delivery of cells in tissue engineering fields.
