利用矮败小麦诱导单倍体的初步研究
A preliminary study on the induction of haploid in Aibai wheat
【摘要】 用化学药剂和远缘花粉处理矮败小麦筛选诱导单倍体的最佳处理组合.结果表明:单独用药剂或玉米花粉授粉诱导效果不理想,其中以0.5mg·L-12,4-D的最好,诱导率为12.7%-13.1%;油菜或玉米花粉授粉后24和48h再经10g·L-1二甲基亚砜(DMSO)+15mg·L-12,4-D处理,以及玉米花粉授粉后再经4mg·L-1BA+10g·L-1DMSO处理,诱导频率较高,达54.8%-77.0%,与其他处理的差异达极显著水平;玉米花粉授粉的诱导效果明显好于油菜花粉的;10g·L-1DMSO+15mg·L-12,4-D处理优于4mg·L-1BA+10g·L-1DMSO处理,两处理诱导率的差异达显著水平.
【Abstract】 Chemicals and pollen from distant species were employed to induce haploid in Aibai wheat. Results showed that treatments of single factors (chemicals or maize pollen) were not efficient. Among them, treatment of 0.5 mg·L-1 2,4D exhibited the highest rate of haploid induction, ranging from 12.7% to 13.1%. As to the composite treatments, rape or maize pollens followed by 10 g·L-1 DMSO+15 mg·L-1 2,4D sprayed at 24 or 48 h after pollination or maize pollen followed by 10 g·L-1 DMSO+4 mg·L-1 BA induced the highest frequencies of haploid, ranging from 54.8% to 77.0%, significantly higher than other treatments. In regard to the factors in the composite treatments, maize pollen was better than rape pollen and 15 mg·L-1 2,4D was better than 4 mg·L-1 BA. The issues of the roles of 2,4D, BA and DMSO and young embryo rescue were discussed.
