新型葡萄糖氧化酶电极与毛细管电泳的联用
A Novel Oxidase Electrode for Determination of Glucose Goupled with Capillary Electrophoresis
【摘要】 采用浸涂法将 1 二茂铁基乙胺和壳聚糖修饰到铂电极表面 ,与葡萄糖氧化酶溶液作用后用戊二醛交联 ,制备壳聚糖固定化葡萄糖氧化酶生物传感器 ,测定葡萄糖在传感器上的伏安曲线 .采用恒电位安培检测法 ,葡萄糖浓度在 0 0 0 2~ 4 0mmol/L范围内与响应电流成线性关系 ,响应时间短 .由于二茂铁衍生物和葡萄糖氧化酶通过包埋、键合等作用固定于壳聚糖膜中 ,不易从电极表面流失 ,电极稳定性较好 .同时尝试将该电极与毛细管电泳联用 ,成功用于人血清中葡萄糖的测定 .
【Abstract】 Ferroceneethylamine and chitosan were modified on a platinum eletrode by dipping method. And then,the modified electrode was sunk is glucose oxidase solution to immobilize the enzyme into the chitosan membrane. A novel glucose oxidase biosensor could be fabricated after it was finally treated with cupling reagent,glutaraldehyde. The glucose V A curve on the target biosensor has been measured. Using amperometric determination method with potential keeping constant,the current response of the sensor is found to be linear with the glucose concentration in the range of 0 002-4 0 mmol/L with fast responese. Since 1 ferroceneehtylamine and glucose oxidase are,through the action of warp and bond,immobilized in the chitosan membrane,being not easy to be lost from electrode surface,the electrode owns high stability. It has been coupled with capillary electrophoresis to determine the glucose in human serum with satisfactory assay results.
