大鼠腹主动脉移植后PDGF和IGF-1表达的测定及其意义

【摘要】 目的 观察血小板衍生生长因子 (PDGF)、胰岛素生长因子 1 (IGF 1 )在移植动脉中的表达规律 ,探讨其在移植动脉硬化中的作用。方法 采用原位大鼠腹主动脉移植模型 ,免疫组织化学SP法测定生长因子的表达。结果 术后 2 1d内膜增厚 ,至 60d异系移植动脉内膜面积比约为同系对照组的 3倍 [术后 60d分别为异系 2 9 2 1 % ,同系 (8 0 3± 0 1 8) % ,P <0 0 1 ]。异系移植组PDGF术后 7d内膜表达至高峰 ,平均评分值 1 51 ,非常显著高于同系移植组 0 2 0 (P <0 0 1 )。IGF 1术后 7d开始上调表达 ,其后 2 1d达高峰 ,平均评分值 0 82 ,非常显著高于同系移植组 0 2 0 (P <0 0 1 ) ,至 60d仍保持高表达 ,平均评分值 0 65。结论 异系移植大鼠腹主动脉增厚明显。IGF 1 ,PDGF通过旁分泌及自分泌的方式促进平滑肌细胞的增殖 ,并由中膜向内膜迁移 ,导致内膜增厚。可能在移植动脉硬化发生过程中发挥重要作用

【Abstract】 Objective To investigate the effects of platelet derived growth factor (PDGF A) and insulin like growth factor 1 (IGF 1) on the development and progress of transplanted arteriosclerosis (TA). Methods The expressions of PDGF A and IGF 1 in the tissues of abdominal aortic allografts/isografts obtained from rat models of in situ transplantation of abdominal aorta were detected with immunohistochemical S P method. Results The ratio of thickening area in tunica intima of the allografts was about threefold more than that in the isografts control 60 d after transplantation [29.21% vs (8.03±0.18)%, P <0.01]. The expression of PDGF reached its peak 7 d after transplantation with an average optical density ( A ) of 1.51, which was significantly higher than 0.20 of isografts ( P <0.01), then declined to baseline on d 21. The expression of IGF 1 began to be upregulated on d 7, and came to the highest on d 21 at an average OD of 0.82,having a significant difference in comparison with isografts group (0.20) ( P <0.01), and still kept highly expressed on d 60 with an average OD of 0.65. Conclusion The intimal thickening is evident in the allografts. IGF 1, PDGF and other growth factors exert autocrine and paracrine effects on vascular smooth muscle cell proliferation, migration from median into intima. Growth factors released during rejection may be involved in accelerating myointimal proliferation. The increment of local PDGF A and IGF 1 synthesis may play important role in the pathogenesis of transplantation arteriosclerosis (TA).
