紫穗槐的离体快速繁殖(英文)
In vitro micropropagation of Amorpha fruticosa
【摘要】 以子叶节为外植体 ,建立起了紫穗槐的快速离体再生系统。经过四周的培养 ,在附加8mg· L- 16 - BA的 MS培养基上能够获得再生频率为 1 0 0 % ,平均每个外植体 5 .2 1个芽点的高效再生植株。以再生植株的茎节为外植体所进行的继代能够在相同的培养基上连续的产生新的不定芽 ,但芽点数要少于起始培养。经过 3周的培养 ,有 82 .5 3 %切下的再生茎段能够在含 2 .0 mg· L- 1IAA的 MS培养基上生根。在所有进行分析过的再生植株中 ,它们的染色体数目都没有发生变异 (2 n=40 )。经过练苗以后 ,再生植株成功地定植于土壤当中并展示了一致的外部形态和生长特性。
【Abstract】 Using cotyledonary nodes as explants,a rapid and efficient method for propagation of Amorpha fruticosa L.has been developed in the present paper.When grown on Murashige and Shoog medium (MS) supplemented with 8 mg·L -1 BAP for adventitious shoots induction,100% explants responded with 5.21 shoots per explant after 4 week’s culture.Explants taken from the in vitro proliferated shoots subsequently produced multiple shoots when cultured on the same medium,but the shoot number per subculture was lower than that of the initiatial culture.82.53% excised shoots were rooted on half-strength MS medium containing 2.0 mg·L -1 IAA after 3 weeks of incubation.The regenerated plantlets possessed normal diploid chromosome number (2n=40) in all the analyzed cells.After acclimatization,all of the rooted plantlets established in soil,exhibiting uniform morphological and growth characteristics.
