菊粉酶酵母菌株的筛选及鉴定
Screening and identification of inulinase-Producing yeast strains
【摘要】 从菊芋周围土壤和实验室保存的菌株中分离筛选产菊粉酶的酵母菌株 ,得到产菊粉酶活力较高的酵母菌 7株。以K .Marxianus为对照 ,对其中 5株未知菌进行菌种鉴定 ,并将它们归列到属。经进一步复筛 ,选定酶活最高的酵母KI0 7,初步研究KI0 7菊粉酶的酶学性质。结果表明该酶的最适作用 pH为 4 .5、最适反应温度为 5 5℃ ,作用于菊粉、蔗糖和棉子糖 ,其I/S、I/R值分别为1/ 2 .80、1/ 2 .89,该酶为外切型菊粉酶。
【Abstract】 We screen inulinase-producing yeast strains from Jerusalem artichoke soil and samples reserved in the laboratory. Seven strains with high activity are obtained. We find the yeast strain KI07, with the highest activity, to be an exoinulinase. The optimal pH and temperature of inulinase from KI07 are 4.5 and 55?℃,respectively .All th inulin, surcrose, and raffinose could be hydrolyzed by inulinase from KI07,with ratios of I/S and I/R for KI07 inulinase being 1/2.80 and 1/2.89.
