克鲁斯假丝酵母植酸酶的筛选及其发酵条件
Screening of C. Krusei WZ-001 phytase and its fermentation condition
【摘要】 筛选出了一株高植酸酶活性的酵母菌株 ,鉴定为克鲁斯假丝酵母C .KruseiWZ 0 0 1。该植酸酶定位于细胞壁上 ,为PHYA型植酸酶。最适作用条件 :pH分别为 3.2和 4 .72 ,温度为 5 5℃ ,保持 6 0 %的酶活性 10min。C .KruseiWZ 0 0 1植酸酶有广泛的底物特异性 ,一定浓度的无机磷对其植酸酶活性有抑制作用。在优化的发酵培养基上 ,最高发酵液酶活性为 1.6 μmol/mL ,干细胞植酸酶活性为 2 80 μmol/ g。
【Abstract】 We screen out a yeast strain producing high activity of phytase, identified as C. Krusei WZ 001. The phytase is located on the cell wall of the type PHYA. Under the optimum condition: 3.2 and 4.72 of pH, 55?℃ of temperature; it keeps 60% of phytase activity for 10?min at 65?℃. The phytase of C. Krusei WZ 001 exhibits a broad substrate specificity and it’s activity is repressed by inorganic phosphorus with certain concentration. In optimum fermentation medium, the highest activity of culture is 1.6?μmol/mL, and the phytase activity of dry cells is 280?μmol/g.
