东北红豆杉内生真菌的分离和筛选
Isolation and Screen of Taxus cuspidata Sieb. et Zucc. Endophytic Fungi
【摘要】 从长白山自然保护区原始森林 2 0 0多年生东北红豆杉 (TasxuscuspidataSiebetZucc.)树皮中分离出 2 1种内生真菌 ,经鉴定 :1种属于子囊菌 ,2 0种属于半知菌。用薄层层析技术 ,测定了各菌种发酵代谢产物 ,经筛选发现 :链格孢 (Alternariasp .,10 11)、葡萄孢 (Botrytissp .,10 14)、散囊菌 (Eurotiumsp .,10 0 1)、曲霉 (Aspergillussp .,10 0 3) 4种内生真菌的代谢产物可与紫杉烷类物质显色剂———香草试液发生较明显的颜色反应。其中 ,链格孢 (Alternariasp .10 11)菌种的代谢产物中有一种含量较大的组分还可与稀碘化铋钾生物碱试剂发生反应 ,经过光谱分析 ,该组分化合物Ⅰ为紫杉烷类二萜Ⅲ型化合物 ,说明该菌能高产紫杉烷类物质。
【Abstract】 Twenty-one endophytic fungi are isolated from the bark of Taxus cuspidata Sieb. et Zucc. growning in the primeval forest of the Changbai mountain natural reserve for more than 200 years. One of the srains is identified as Ascomycetes and the others are Fungi imperfecti. Through thin layer chromatography (TLC) to determine the liquid fermentation metabolites of the strains, four strains, Alternaria sp., Botrytis sp., Eurotium sp., Aspergillus sp. are screened, whose metabolites can react positively with the vanillic aldehyde, one special taxoid developer. Among the four, Alternaria sp. 1011 can produce one compound in a large amounts, which can positively react with one alkaloids developer--Bismuth potassium iodide. The compound is identified as taxoids type Ⅲ through kinds of spectrum analysis. This demonstrates that Alternaria sp. 1011 can highly produce one kind of taxoids.
