GA20-氧化酶基因转化豆科牧草百脉根的研究
<title>Transformation of Leguminous forage Lotus corniculatus by Agrobacterium mediated GA 20oxidase gene
【摘要】 以豆科植物百脉根子叶为转化受体,通过根癌农杆菌介导方法将外源目的基因GA20-氧化酶(GA20-oxidase)和NPTII基因导入,经筛选分化、再生,得到具有卡那霉素抗性的转基因植物。在卡那霉素筛选浓度的选择中,25mg/L是百脉根较为适宜的筛选浓度。头孢霉素对农杆菌的抑制作用好于羧卞霉素。转基因植物移栽大田后生长良好。PCR检测证明外源目的基因已整合到百脉根基因组中。
【Abstract】 <Abstrcat> Transgenic plant of Lotus corniculatus L. was obtained by Agrobacteriummediated transformation, utilizing a binary vector containing GA 20oxidase gene and Kanamycin resistance gene NPTII. Transgenic plants grew well after transferring to greenhouse. PCR analysis confirmed that GA 20oxidase gene and NPTII gene were integrated into the Lotus corniculatus genome.
