壳聚糖固定化木瓜蛋白酶的制备及性质

【摘要】 以壳聚糖为载体 ,戊二醛为交联剂制备固定化木瓜蛋白酶 .研究了固定化时间、温度、pH值、给酶量和戊二醛浓度对固定化木瓜蛋白酶活力的影响 ,结果表明 :木瓜蛋白酶最佳固定化条件是给酶量为每克载体 10mg ,戊二醛浓度0 .5 % ,pH7.5 ,室温下 (2 5℃ )反应 10h .所制备的固定化木瓜蛋白酶的最适pH8.0 ,最适温度 70℃ ,与溶液酶相比 ,固定化酶的热稳定性显著提高

【Abstract】 Papain has been immobilized on chitosan with glutaraldehyde as crosslinking agent.The factors involving with the activity of immobilized ,such as reaction time,temperature,pH,amount of papain and concentration of glutaraldehyde,were studied.The results showed that the optimum condition for immobilization were as follows:the load of enzyme was 10mg/g carrier,0.5 solution of glutaraldehyde,pH7.5,the reaction time was 10h at 25℃.Stability of immobilized Papain to temperature was increased obviously compared with the soluble papain.
