伊文思蓝脂质体的制备及其在小鼠体内的分布

【摘要】 目的 :研究伊文思蓝脂质体在小鼠体内的组织分布状况 ,为研究脑靶向脂质体奠定基础。方法 :用反相蒸发法制备伊文思蓝脂质体 ,用凝胶柱分离的方法测得脂质体的包封率为 2 5 .0 7% ,比较了伊文思蓝脂质体和伊文思蓝水溶液在小鼠体内的组织分布状况。结果 :磷脂与胆固醇的比例对伊文思蓝脂质体的包封率有较大的影响 ;尾静脉注射后 2~ 4h时脂质体组在脑中伊文思蓝的浓度高于水溶液组。结论 :伊文思蓝可作为检测脂质体跨血脑屏障的模型药物 ,脂质体对药物跨血脑屏障有一定的促进作用

【Abstract】 Objective: To study the distribution of liposomes in mice and to improve the drug delivery system across the blood brain barrier. Methods: Liposomes containing Evens blue were prepared by the method of REV and separated by the Sephradex G-50. The encapsulation percentage of Liposomes containing Evens blue was 25.07%. The distribution of the Evens blue solution and Liposomes containing Evens blue were both studied. Results: The ratio between phospholipids and cholesterol had influence on the incorporation rate of Liposomes containing Evens blue. The concentration of Evens blue in the mouse brain 2-4 h after the liposomes containing Evens blue were injected was greater than that of the Evens blue solution. Conclusion: Evens blue is a good marker to determine the integrity of the blood brain barrier. Liposomes can improve the ability of the drug across the blood brain barrier. A new drug delivery system across the blood brain barrier is offered.
