骨髓间质干细胞的体内成骨特性

【摘要】 目的 :观察大白鼠骨髓间质干细胞 (bonemarrowmesenchymalstemcells,BMMSCs)的体内成骨方式 ,研究新生骨组织的生物学特点、形态及骨基蛋白的表达。方法 :取 2月龄SD大鼠的胫骨及股骨骨髓进行培养 ,体外培养的BMMSCs经成骨诱导剂诱导后分别与羟基磷灰石磷酸三钙陶瓷 (hydroxyapatite tricalciumphosphateceram ics,HA TCP)、羟基磷灰石磷酸三钙陶瓷加藻酸钠 (alginatesodium ,AS)移植入BALB C裸鼠皮下。 2个月后取出种植物 ,组织切片观察成骨方式及成骨类型。用免疫组化法对新生骨骨基质蛋白的表达进行检测。结果 :BMMSCs+HA TCP与BMMSCs +HA TCP +AS均为膜性骨发生 ,2个月内所成的骨为层板骨。此方法所成骨的骨涎蛋白、骨桥蛋白、Ⅰ型胶原均有表达。结论 :BMMSCs通过HA TCP以膜性骨发生方式成骨 ,2个月内成的骨为层板骨。所成的骨与胚胎发育的骨在骨涎蛋白、骨桥蛋白、Ⅰ型胶原的表达无区别。

【Abstract】 Objective: To observe osteogenesis type of rat bone marrow mesenchymal stem cells (BMMSCs) in vivo and study the characteristics, morphology and expression of bone matrix protein of new bone. Methods: BMMSCs were collected from bone marrow of tibiae and femurs of SD rats. Culture of BMMSCs induced by osteogenic medium was seeded on hydroxyapatite/tricalcium phosphate ceramics (HA/TCP). Thereafter they were transplanted subcutaneously into immunodeficient mice. HA/TCP was also implanted with BMMSCs and alginic sodium. Two months later samples were examined histologically to see whether new bone had formed. Samples were also tested immunohistochemically whether bone matrix proteins were expressed. Results: Intramembranous ossification occurred both in HA/TCP+BMMSCs group and in HA/TCP+BMMSCs+alginic sodium group. The newly formed bone was all lamella bone. Bone sialoprotein, osteopontin and collagen Ⅰ were all expressed in new bone. Conclusion: In HA/TCP, BMMSCs form bone in the way of intramembranous ossification. Within 2 months only lamella bone can be observed. Bone sialoprotein, osteopontin and collagen Ⅰ are all expressed.
