活血渗湿方对实验性肝纤维化大鼠胶原纤维合成与降解的影响

【摘要】 目的:研究活血渗湿方对胶原纤维增生的影响。方法:采用四氯化碳实验性肝纤维化模型并以活血渗湿方干预。与克隆伽玛对照,测定肝功能、血清透明质酸,并通过光镜观察肝组织病理变化,用苦味酸-天狼红染色和氨银-Masson网织纤维染色观察Ⅰ、Ⅲ型胶原,免疫组织化学法检测Ⅳ型胶原及组织金属蛋白酶抑制因子(TIMP-1)的表达。结果:活血渗湿方组较模型组比较,其特点在于:(1)肝功能改善;(2)血清透明质酸显著降低;(3)Ⅰ、Ⅲ、Ⅳ型胶原纤维增生得以明显改善;(4)TIMP-1表达减少。结论:活血渗湿方可通过改善胶原纤维的增生达到抗肝纤维化作用。

【Abstract】 In order to study the effect of promoting the blood flow and resolving dampness recipe on the collagen fiberhy-perplasia classic CCL4 - induced experimental hepatofibrosis model was used and pretreated by promoting the blood flow and resolving the dampness recipe contrasted with clone Gama to test liver function, hyaluronic acid in serum and observe pathological alteration and fibrosis degree of liver tissue. Through optomicroscope and observe collagen Ⅰ、3Ⅲthrough trinitrophenol - Sinus Red and ammonia - Argentum mason dyeing, test collagen Ⅳ and expression of TTMP - I by im-munohistochemistry. The results: Compared with the model group liver function ameliorated, serum hyaluronic acid markedly reduced, collagen Ⅰ、Ⅲ、Ⅳ fibrosis hyperplasia obviously improved and the expression of TIMP decreased in promoting blood flow and resolving the dampness recipe group. Conclusion: The recipe of promoting the blood flow and resolving the dampness can attain to anti - fibrosis function by ameliorating the hyperplasia of collagen fiber.
