蝎毒抗癌多肽对人白血病细胞生长的影响

【摘要】 目的:观察蝎毒抗癌多肽(APBMV)对体外培养的人早幼粒白血病细胞株HL-60生长的影响。方法:细胞毒性实验(MTT法)和生长抑制实验。结果:APBMV可使HL-60代谢MTT的能力显著低于对照组,其IC50值为10.74μg/ml;在适当浓度,APBMV可显著抑制HL-60细胞的生长(P<0.01),剂量效应与时间效应关系明显,24h、48h和96h的IC50值分别为19.41μg/ml、9.90μg/ml、11.41μg/ml,且24h与48h间、48h与96h间效应相比,均有显著相关关系(r=0.9660,P<0.05;r=0.9940,P<0.01);APBMV对HL-60细胞的生长抑制作用以48h为著,96h时效应不增强。结论:APBMV是东亚钳蝎蝎毒中有效低毒的抗肿瘤成分,可显著抑制白血病细胞的生长。

【Abstract】 Cytotoxic assay (MTT colorimetric method) and growth-inhibiting test was used to observe the growth influence of antineoplastic polypeptide from Buthus martensii venom (APBMV) on the cultured human promyelocytic leukemia cells HL-60. The result shows that APBMV could induce HL-60 metabolizing MTT a-bility to dramatically decrease, and IC50 was 10.74μg/ml; at appropriate concentration, APBMV could obviously inhibit HL-60 cell growth (P<0.01); there was evident correlations in dosage-response and time-response; the IC50 at 24h, 48h and 96h were 19.41μg/ml、 9.90μg/ml、11.41μg/ml respectively; there were instinct correlations in inhibiting effect between 24h and 48h, or 48h and 96h (r = 0.9660, P < 0.05; r = 0.9940, P < 0.01) ; the inhibiting effect of APBMV on growth of HL - 60 cells was stronger at 48h interval. The conclusion is that APBMV was effective and low toxic antineoplastic component in the venom of Buthus Martensii Scorpion and could 4notably inhibit the growth of leukemia cells.
