益气温阳护卫汤对哮喘豚鼠BALF Th1、Th2细胞因子产生及调节的影响
Effect of Yiqiwenyanghuwei Decoction on Th1 and Th2 Cytokines Production and Regulation in BALF in Asthma Guinea-pigs
【摘要】 目的:观察益气温阳护卫汤对哮喘豚鼠支气管肺泡灌洗液(BALF)Th1、Th2细胞因子产生及调节影响。方法:将豚鼠随机分为正常组、哮喘组、地塞米松组、玉屏风散组及益气温阳护卫汤低剂量组、中剂量组、高剂量组;于诱喘后24小时采集标本,并采用双抗体夹心ELISA法测定Th1细胞分泌的细胞因子干扰素-γ(IFN-γ);Th2细胞分泌的细胞因子白细胞介素-4,5(IL-4,IL-5);采用比色法测定各组BALF一氧化氮(NO)含量。结果:益气温阳护卫汤各剂量组均能提高IFN-γ水平,降低IL-4,IL-5水平。中、高剂量组与地塞米松组IL-4水平无明显差异(P>0.05);高剂量组IL-5水平虽低于地塞米松组,但无明显差异(P>0.05),与其他各组均有差异(P<0.05)。高剂量组IFN-γ浓度与其他各组比较有显著性差异(P<0.05),中、低剂量组、玉屏风散组与地塞米松组比较无显著性差异(P>0.05),但这四组均高于哮喘组(P<0.05)。BALF NO水平,以高剂量组最低,但与中剂量组及地塞米松组比较无显著性差异(P>0.05),三者与其他组比较有显著性差异(P<0.05)。结论:益气温阳护卫汤可以促进Th1细胞分泌IFN-γ,抑制Th2细胞分泌IL-4,IL-5;调节Th1、Th2细胞因子产生的可能有效途径之一,是通过降低NO水平实现。
【Abstract】 To investigate the effect of Yiqiwenyanghuwei decoction on Th1 and Th2 cytokines production and modulation in bronchoalveolar lavage fluids (BALF) in asthma guinea - pigs. Methods: The guinea-pigs were randomly divided into seven groups, the normal control group, the asthma group, the dexmethaone group, the Yu-pingfeng powder group and the three different dose’s groups of Yiqiwenyanghuwei decoction. The guinea-pigs’ samples of BALF were collected 24h after asthma provoked. The levels of cytokines such as interleukin - 4 (IL-4), interleukin - 5 (IL - 5), which are secreted by Th2, and interferon-γ (IFN -γ) which is secreted by Th1 were measured by enzyme linked immunosorbent assay (ELISA) ; The levels of nitric oxide (NO) were measured by photometry. Results: The different dose’s groups of Yiqiwenyanghuwei decoction all could increase the levels of IFN - γand lower IL - 4, IL - 5. There were not significant difference among the middle -dose’s group, the high-dose’s group, and the dexmethaone group in IL-4 levels (P>0.05); the level of IL-5 in the high- dose’s group were lower than that in the dexmethaone group, but there were not significant difference (P > 0.05 ) , significant difference compared with the rest ( P < 0.05). The levels of IFN - γin the high- dose’s group were higher than those in other groups (P>0.05) . the levels of IFN - γamong the middle-dose’ group, the low - dose’s group, the Yupingfeng powder group and the dexmethaone group had no significant difference (P>0.05), but all were significantly higher than that in asthma group. The levels of NO in the high-does’s group were lowest, but had no significant difference compared with the middle-dose’s group and the dexmethaone group (P > 0.05), and significant difference compared with the other groups ( P < 0.05). Conclusion: Yiqiwenyanghuwei decoction could promote Th1 to the secret IFN - 7, and inhibit Th2 to the secret IL - 4, IL - 5; one of the effective ways of regulating the cytokines production of Th1 and Th2 may be through lessening the levels of NO.
