α-、β-雌二醇和中药有效成分ZDY101对CHOm2转基因细胞M2受体的影响
A Comparative Study on the Effects of α-,β-estradiol and an ZDY101, an Active Component of Yin Tonic on M2 Receptor of CHOm2 Transgenic Cells
【摘要】 本文用细胞培养技术 ,对比观察了α -雌二醇 (α -E2 )、β -雌二醇 (β -E2 )和滋阴药有效成分ZDY10 1对转染M2 受体基因的CHO细胞M2 受体密度的影响。在CHOm2 转基因细胞培养 2 4h后 ,加入不同浓度的α -E2 、β -E2 或ZDY10 1。继续培养 4 8h后收集细胞 ,用3 H -QNB单点法测定M2 受体密度。发现α-E2 及 β -E2 在浓度为 1× 10 -5mol/L时 ,M2 受体密度明显高于对照组 ,与相同浓度的ZDY10 1作用相仿。以上结果提示 :由于α -E2 及β-E2 对M2 受体密度的作用相似 ,所以E2 对M2 受体的调整作用 ,其受体后的信息传递机制很可能与对雌性器官的调整作用是不同的 ;由于ZDY10 1与E2 结构上和调节M2 受体的作用有类似处 ,它和E2 及E2 受体的关系值得深入研究
【Abstract】 This paper aimed at the comparative observation on the effect of α-estradiol(α-E 2), β-estradiol(β-E 2) and ZDY101, an active component of Yin Tonic, in regulating the density of M 2 receptors of CHOm 2 transgenic cells. After the cells were cultured for 24h, α-E 2, β-E 2 or ZDY101 was added to the culture medium, the final concentration was 10 5 mol/L or lower. After a further culture for 48h, the cells were collected and measured for their M 2 receptor density using single site 3H-QNB binding assay. It was found that the M 2 receptor density was elevated by 1×10 -5 mol/L of α-E 2 or β-E 2 to similar extent but not by lower concentration of either E 2. Since α-E 2 showed much weaker effect in regulating sexual functions than β-E 2, it is likely that both E 2 regulate the M 2 receptors by signal pathways different from regulating sexual functions. It was also found that 1×10 -5 mol/L of ZDY101 raised the M 2 receptor density to similar extent as E 2. Since the chemical structure of ZDY101 was similar to E 2, whether ZDY101 raised the M 2 receptor and hence improved the learning and memory of Alzheimer′s patients through E 2 receptor is an interesting problem and is worth of further study.
