天然生物膜对长波紫外线氧化损伤Hela上皮细胞的保护作用
PROTECTIVE EFFECTS OF NATURAL BIOLOGICAL MEMBRANE ON Hela CELLS DAMAGED BY UVA
【摘要】 ①目的 探讨天然生物膜对长波紫外线 (UVA)辐射所致Hela上皮细胞氧化损伤的保护作用及其机制。②方法 建立UVA对Hela上皮细胞氧化损伤模型。Hela上皮细胞实验设计分为 4组 ,未损伤组、UVA损伤对照组、UVA +10 g/L生物膜组、UVA +10 g/L维生素C组。MTT法测定细胞活性 ;流式细胞仪测定细胞的凋亡和死亡率 ;酶法测定抗氧化酶活性和MDA的含量及总抗氧化能力。③结果 离体条件下生物膜能显著增加Hela上皮细胞的活性 ,减少细胞的凋亡和死亡率 ;提高细胞上清液中谷胱甘肽过氧化物酶 (GSH Px)、过氧化氢酶(CAT)、超氧化物歧化酶 (SOD)的活性 ,降低丙二醛 (MDA)的含量 ,且呈量效关系。④结论 生物膜在体外有抗UVA氧化损伤的作用。其作用机制可能是通过提高抗氧化酶含量、清除自由基等 ,以减轻细胞损伤而发挥其保护作用的。
【Abstract】 Objective\ To assess the protective effects of natural biological membrane (NBM ) on Hela epithelia cells damaged by ultraviolet A(UVA).\ Methods\ The oxidative damage models of Hela epithelia cells irradiated by UVA were established. The experiment was carriedout in 4 groups: Undamaged Group, UVA damaged Control Group, UVA+10 g/L Biological Membrane Group, and UVA+10 g/L Vitamin C Group. The cell activities were tested by MTT method; the rates of apoptosis and mortality of the cells were tested by flow cytometry; the concentration of MDA and the activities of GSH Px, SOD, and CAT in the supernatant of cells were determined by biochemical technique. \ Results\ The experiment indicated that, in vitro, NBM could enhance the activities of Hela cells, decrease apoptosis and mortality, increase the activities of GSH Px, SOD, CAT and decrease the content of MDA in the supernatant of Hela cells. \ Conclusion\ The NBM has the protective effects on the Hela cells irradiated by UVA. The mechanism of this protective effects of NBM may be mediated by eliminating the free radicals and increasing the activities of antioxidative enzymes.
