柴胡加龙骨牡蛎汤对戊四唑点燃癫痫大鼠海马内cAMP、cGMP含量的影响

【摘要】 目的 :探讨戊四唑 (PTZ)点燃癫痫模型大鼠海马内cAMP、cGMP的变化及柴胡加龙骨牡蛎汤对其影响。方法 :成年SD大鼠 4 5只随机分为 3组(每组 1 5只 ) :正常对照组、癫痫模型组、柴胡加龙骨牡蛎汤组。采用放射免疫法测定海马内cAMP、cGMP含量。结果 :癫痫模型组海马内cAMP、cGMP含量较正常对照组明显增加 (P <0 .0 5或P <0 .0 1 ) ,cAMP/cGMP比值较正常对照组降低(P <0 .0 1 ) ;经柴胡加龙骨牡蛎汤治疗后cAMP有所增加 ,而cGMP、cAMP/cGMP值恢复正常。结论 :柴胡加龙骨牡蛎汤的抗癫痫作用与海马内cAMP、cGMP含量变化有关。

【Abstract】 Objective:To observe the changes of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monphosphate (cGMP) level in the hippocampus of pentylene tetrazole (PTZ) kindled epilepsy model rats, and explore the effects of Chaihu Jia Longgu Muli (CJLM) decoction. Method: 45 adult SD rats were randomly divided into three groups (15 rats in each group), namely, control group, epilepsy model group, CJLM decoction treated group. The levels of cAMP and cGMP in the hippocampus homogenate were assayed by radioimmunoassay.Result:The levels of cAMP and cGMP in the hippocampus of epilepsy model group were significantly decreased in comparison with those of control group (P<0.05 or P<0.01). The ratio of cAMP to cGMP was markedly decreased compared with that of control group (P<0.01). After treated with CJLM decoction, the cAMP level was significantly increased, and the ratio of cAMP to cGMP was restored to normal.Conclusion: The therapeutic effects of CJLM decoction on epilepsy are related to the changes of cAMP and cGMP levels.
