反相高效液相色谱法测定全血中环孢霉素A

【摘要】 目的 建立一种全新的反相高效液相色谱法 (RP HPLC法 ) ,用于测定全血中环孢霉素A(CsA)。方法 在甲醇 -乙腈-水体系中 ,研究影响CsA测定的诸条件 :流动相的组成、柱温的降低及内标物的选择等 ;提出RP HPLC法测定CsA的新方法 :于C18柱上 ,用含 0 0 0 2 5mol·L-1Li2 SO4 的甲醇 -乙腈 -水 (16∶6 6∶18,V/V)为流动相 ,以环孢霉素C(CsC)为内标物 ,在5 0℃柱温下 ,于 2 18nm处检测。结果 CsA线性范围 2 5~ 16 0 0 μg·L-1,最低检测浓度 10 μg·L-1,回收率 10 0 2 1%,日间和日内变异系数均小于 6 0 %;全血中CsA线性范围 2 5~ 16 0 0 μg·L-1,最低检测浓度 10 μg·L-1,回收率 90 14%,日内和日间变异均小于 9 0 %。结论 本法具有简便、灵敏、快速、线性范围宽、稳定性好、柱温低的特点 ,可广泛用于临床血药浓度监测。

【Abstract】 AIM To develop a new RP-HPLC method for determination of Cyclosporine A(CsA) in whole blood. METHODS The effects of the composition of mobile phase, the reduction of analytical column temperature and the choice of internal standard on the determination of CsA were investigated through the systems including methanol, acetonitrile and water. A new method of RP-HPLC determination of CsA was advanced: a RP-C 18 was used as the stationary phase with a mobile phase of methanol-acetonitrile-water( 0.002 5 mol·L -1 Li 2SO 4 16∶66∶18,V/V). UV detection was performed at 218 nm. The oven temperature was 50℃. Cyclosporine C was used as the internal standard. RESULTS In whole blood the liner range was 25~ 1 600 μg·L -1. The mininal detectable concentration of this method was 10 μg·L -1. The recovery was 90.14%. The RSD% was less than 9.0%. CONCLUSION The method has the following features:convenience, sensitivity, rapidity, wide liner range, good stability and low oven temperature.It could be widely used to determine the concentration of CsA in whole blood.
