端粒酶hTERT在非小细胞肺癌的表达

【摘要】 目的 探讨非小细胞肺癌 (NSCLC)组织中端粒酶逆转录酶 (hTERT)表达的分布及其和端粒酶活性表达的关系。方法  35例原发性非小细胞肺癌和 10例正常肺组织冰冻切片经抗hTERT抗体免疫组化染色 ,观察hTERT表达的分布。同时测定标本的端粒酶活性 ,分析两者的相关性。结果  35例肿瘤标本中 ,用免疫组化分析hTERT表达呈阳性2 7例 ,镜下观察阳性信号为棕黄色颗粒 ,主要分布于癌细胞的细胞质。用TRAP法测定 ,端粒酶活性表达呈阳性 30例 ,hTERT呈阳性表达的 2 7例标本中端粒酶活性也呈阳性的为2 2例。 10例正常组织二者均为阴性。结论 hTERT仅在肺癌细胞中的表达 ,且与端粒酶活性的表达相一致。

【Abstract】 Objective To investigate the distribution of human telomerase reverse transcriptase expression in non small cell lung carcinoma (NSCLC) and the relationship with telomerase activity. Methods 35 NSCLC and 10 nomal lung tissues were analysized by immuno histochemistry with the anti hTERT antibody for the hTERT expression and telomeric repeat amplyfication protocol (TRAP) assay for telomerase activity. Results 27 tumors were graded as positive for hTERT expression by immuno histochemistry and 30 for telomerase activity by TRAP assay, A close relation between them was observed. All the normal tissue samples were negative in both cases. In tumors that were positively stained, signals were localized specifically to the cytoplasm of the carcinoma cells. Conclusion In NSCLC, hTERT is specifically expressed in carcinoma cells and corellatted with the activation of telomerase activity.$$$$
