HBVx转基因小鼠肝组织基因表达谱的微阵列研究
THE STUDY OF HBVx GENE AND AFLATOXIN B1 ON cDNA ARRAY OF LIVER IN TRANSGENIC MICE WITH HBVx GENE
【摘要】 目的 研究乙肝病毒 (HBVx)和黄曲霉毒素(AFB1)在诱发肝癌中的分子机理。方法 用AtlasTM cDNA表达阵列方法比较研究HBVx基因转基因小鼠HBVx基因整合 (+)和 (- )的小鼠肝组织 ,以及转基因小鼠经AFB1一次攻击的肝癌组织的基因表达谱的差异。结果 经HBVx基因整合 (+)的小鼠肝组织的DNA修复基因 ,药物代谢酶基因以及应激反应的效应因子的cDNA基因表达谱显示了明显的变化。实验结果提示出现变异的 30个cDNA基因表达谱中 2 9个 (96 .7% )与X基因的整合相关及 17个 (5 6 .7% )与AFB1的攻击有关。结论 HBV对宿主的感染 ,可能通过改变DNA修复系统和药物代谢酶系统来影响机体对AFB1的致癌敏感性 ,从而提高AFB1对肝组织的致癌效应
【Abstract】 Objective To investigate the molecular mechanism of interac tion between HBV and AFB 1 in HBVx transgenic mice.Methods Using Atlas TM cDNA array to analyze the mice liver tissues with/without HB Vx integration and treated untreated with AFB 1.Results HBVx integration induced the alteration of cDNA expression. Among them, 29 out o f 30(96.7%) expression of cDNA were related to HBVx integration, while 17 out of 30(56.7%) were associated with AFB 1. And the alteration of cDNA expression m ainly occurred in DNA repair, drug & xenobiotic metabolism and stress response regulators.Conclusion These results indica ted that infection of HBV enhanced susceptibility host to hepatocarcinogenic act ion of AFB 1 by altering DNA repair system and drug metabolism system.
