γδT细胞的细胞毒活性研究

【摘要】 目的 探讨选择性扩增人γδT细胞的方法及其细胞毒活性。方法 用耐热性结核杆菌低分子多肽抗原 (Mtb)刺激正常人PBMC ,并用磁珠阳性分选法获得高纯度的γδT细胞 ,通过MTT试验观察γδT细胞对人红白细胞白血病细胞株K5 6 2的细胞毒活性。结果 Mtb刺激及磁珠分选后的γδT细胞可达 73 %和 98 0 5 % ,效靶细胞比例为 10 :1时其对K5 6 2细胞的杀伤率分别为 5 9%和 78 5 %。结论 Mtb刺激及磁珠分选所获得的高纯度γδT细胞可介导对K5 6 2细胞的高效细胞毒活性

【Abstract】 Objective To study the specific expansion method and cytotoxic activity of human γδT lymphocytes. Methods The heat resistant peptide antigen derived from mycobacterium tuberculosis (Mtb) was applied to stimulate the peripheral blood mononuclear lymphocytes from healthy donors. γδT lymphocytes were coated by TCR γ/δ hapten antibody in combination with MACS anti hapten MicroBeads FITC, and were separated with a LS + positive selection MACS separator. The cytotoxic activities of γδT lymphocytes were monitored by MTT colorimetric assay. Results The purities of the γδT lymphocytes stimulated by Mtb and sorted by MACS were up to 73 % and 98 05 % respectively. The cytotoxic activities of these γδT lymphocytes to K562 cell line were 59 % and 78.5 % when effector/target cell ratio was 10:1. Conclusion The γδT lymphocytes stimulated by Mtb and sorted by MACS mediated strong cytotoxic activity to K562 cell line.
