【摘要】 采用MTT法和流氏细胞仪技术 ,研究了高三尖杉酯碱对HL -60细胞和QGY770 3细胞的存活率和细胞周期分布影响。结果表明 ,高三尖杉酯碱对HL -60细胞的IC50 是 1 .2 8umol.L-1,对QGY770 3细胞的IC50 是 0 .5 5umol.L-1。高三尖杉酯碱处理HL -60细胞 2 4h后 ,G0 -G1期细胞分布有所增加 ,G2 -M期和S期细胞分布有所降低 ;高三尖杉酯碱处理QGY770 3细胞 2 4h后 ,G2 -M期分布略有降低 ,G0 -G1期细胞分布略有增加 ;高三尖杉酯碱处理HL -60细胞2 4h后 ,G0 -G1期峰前有亚二倍体峰出现 ,说明高三尖杉酯碱在所用剂量下能够诱导HL -60细胞发生凋亡。更多还原

【Abstract】 The cell livability and cell cycle of HL-60 cells and QGY7703 cells treated by homoharringtonine was observed by MTT colorimetry and flow cytometry. We found that the IC 50 of HL-60 cells to homoharringtonine is 1.28 umol.L -1 and the IC 50 of QGY7703 cells to homoharringtonine is 0.55 umol.L -1 . After treated by homoharringtonine for 24 hours, G 0 -G 1 period of HL-60 cells increased, G 2 -M and S period of HL-60 cells decreased; G 2 -M period of QGY7703 cells decreased a little, G 0 -G 1 period of QGY7703 cells increased a little; The appearance of subdiploid peak proved the apoptosis inducing activity of homoharringtonine when it treated HL-60 cells for 24 hours.更多还原
