苯白血病患者血清谷胱甘肽转移酶M1(GSTμ)亚型酶活性分析

【摘要】 目的探讨GST μ亚型酶活性的高低与苯白血病之间的关系。方法采用三氯醋酸(TCA)同步终止法对 34例苯白血病患者血清谷胱甘肽转移酶 M1(GSTμ)亚型酶活性进行了测定。结果苯白血病患者 GST μ亚型酶活性 较白血病患者和正常人群低,而白血病患者GST μ亚型酶活性与对照组差异无显著性。在GSTμ缺失的情况下,无 GSTμ亚型酶活性;但在GSTμ基因携带的情况下,有的样品中GSTμ 亚型酶活性极低,甚至为零。若GST μ基因携 带者的μ亚型酶活性下降到一定的量值,则可能发生苯白血病或白血病,即白血病的发生主要取决于GSTμ亚型酶活 性;而苯白血病的发生是由 GSTμ基因缺失率增加和 GSTμ基因携带者的 GST亚型酶活性缺失或极低这两个条件来 决定的。GSTμ亚型酶活性缺失或极低可能是苯致白血病的原因。结论GSTμ亚型酶活性改变可能是苯致白血病的 重要因素之一。

【Abstract】 Objective To explore the relationship between the activities of glutathione s-transferase μ (GSTM1) isoenzyme and leukemia exposed to benzene. Methods The activities of GSTμ isoenzyme in 34 leukemia patients exposed to benzene were determined by the in-phase terminational method of trichloroacetic acid (TCA). Results The activities of GSTμ isoenzyme in leukemia patients exposed to benzene is lower than that of leukemia patients and the controls. GSTμ gene deletion is an important factor affecting GSTase activities. General speaking, GSTμ gene deletion has no GSTμ isoenzyme activity, but under the GSTμ gene circumstance, sometimes there is no GSTμ isoenzyme activity because of no expression of GSTμ or a lower expression of the enzyme from leukemia patients exposed to benzene. Conclusion Leukemia patients exposed to benzene correlates with GSTμ isoenzyme activities and the GSTμ isoenzyme activity is one of the important factors for susceptibility to leukemia patients exposed to benzene.
