普罗布考对冠状动脉球囊损伤后管壁原癌基因c-myc表达的影响

【摘要】 目的 :通过建立猪冠状动脉球囊损伤模型 ,观察普罗布考对血管内膜增生及管壁原癌基因c myc表达的影响。方法 :选用雌性NewYorkshire良种猪 19头 ,随机分为 2组 ,对照组 11头 ,普罗布考组 8头 ,均行冠状动脉球囊扩张术 ,术后喂饲 6周 ,重复冠状动脉造影后处死动物。普罗布考组于术前 4周给予普罗布考 1g ,bid ,至实验结束。常规病理制片 ,观察形态学变化 ,采用RT PCR的方法检测c mycmRNA的表达强度。 结果 :对照组损伤动脉内膜可见大量平滑肌细胞增生 ,呈偏心性增厚 ,普罗布考组内膜增厚程度明显减低。对照组c mycmRNA相对表达强度为 (2 .73± 0 .5 1) ,明显高于正常动脉 [(0 .82± 0 .0 9) ,P <0 .0 5 ],普罗布考组c mycmRNA相对表达强度为 (1.44± 0 .2 5 ) ,较对照组减弱 (P <0 .0 5 ) ,但仍高于正常动脉 (P <0 .0 5 )。结论 :普罗布考具有抑制冠状动脉球囊损伤后c myc表达和内膜增生的作用

【Abstract】 Objective:To observe the effect of probucol on c myc genes and intimal hyperlasia in pig model.Methods:19 female New Yorkshire pigs were divided to receive 1g probucol twice daily ( n = 8) or control therapy ( n =11) for 4 weeks.Then all the pigs were undergone percutaneous transluminal coronary angioplasty(PTCA) and after that probucol and control treatment were continued for another 6 weeks.All the animals were killed for histopathologic examination.RT PCR method was adopted for detection of c myc genes expression.Results:A large quantity of smooth muscle hyperplasia in injured intima were found in control group,but it was significantly fewer in active group.The c myc genes expression in control group [(2.73±0.51)]was obviously higher than that of normal artery [(0.82±0.09), P <0.05],and the c myc genes expression in active group [(1.44±0.25), P < 0.05] was reduced but still higher than that of normal artery( P <0.05).Conclusion:Probucol can decrease the c myc genes expression and intimal hyperplasia in pigs after PTCA.
