【摘要】 观察耳蜗外毛细胞发生凋亡时听力改变情况。方法 :对 2 0只豚鼠药物造模 ,诱发耳蜗外毛细胞发生凋亡。应用TUNEL技术观察凋亡表达 ,测试ABR阈值观察听力变化。结果 :应用丁胺卡那霉素 1天即可诱发豚鼠耳蜗外毛细胞发生凋亡 ,连续应用 3d ,耳蜗外毛细胞凋亡呈强阳性表达 ,但ABR阈值无明显改变 ;随着用药时间延长 ,凋亡细胞数目增加 ,甚至出现部分毛细胞缺失现象 ,此时ABR阈值明显升高。结论 :耳蜗外毛细胞发生凋亡早期对豚鼠听力无明显影响 ,随着耳毒性药物应用时间延长 ,豚鼠ABR阈值升高可能存在两种原因 :毛细胞凋亡或毛细胞坏死。更多还原

【Abstract】 Objective:To investigate the influence of hearing loss of outer hair cell apoptosis in guinea pig cochlea.Methods:Apoptosis of outer hair cell in 20 guinea pigs was induced by drugs,TdT mediated Peroxidase dUTP nick end labeling (TUNEL) techniques were used to detect the apoptosis and measure the ABR threshold.Results:the morphological changes of apoptosis were noted at 1d with amikacin,there were special morphological changes at 3ds with drugs,but ABR has no bvious change.There was an elevation of ABR at 6 successive ds with drugs.Conclusions:There was no considerable chenges in guinea pigs hearing at the early time of apoptosis.Two reasons may contribute to the elevation of ARB threshold at 6 successive with drugs:1) outer hair cell apoptosis;2) outer harir cell necrosis.更多还原
