鸡传染性贫血病毒分离株凋亡素基因的克隆及序列测定
Cloning and sequencing of apoptin gene of chicken infectious anemia virus isolate
【摘要】 使用 PCR技术扩增出鸡传染性贫血病毒分离株 SR43 的凋亡素基因 ,大小为 40 0 bp左右 ,将该片断克隆到 p Bluescript- ks载体中 ,获得含分离株 SR43 凋亡素基因的重组质粒。序列分析表明 ,该分离株凋亡素基因全长36 3bp,编码 12 1个氨基酸 ,与德国标准株 Cux- 1具有高度的同源性 ,仅有 1个氨基酸的差异 ,发生在 116位 ,该分离株为 R(精氨酸 ) ,而 Cux- 1为 k(赖氨酸 ) ,认为 116位氨基酸的改变可能与毒株的毒力有关
【Abstract】 An apoptin gene (VP3 gene) from chicken infectious anemia virus (CIAV) isolate SR 43 ,with the length of 363bp, was amplified by PCR method. The fragment was inserted into vector pBluescript ks and a recombinant plasmid was obtained. The gene was sequenced and its amino acid sequence deduced. It contained 363bp, encoding for 121 amino acid residues. The result showed that it had a highly homology with that of standard CIAV strain Cux 1 from Germany, there is only one amino acid rasidue difference between them (R for isolate SR 43 , K for strain Cux 1 at the position 116), it suggest that may be related to the virulence of different CIAV strains.
