【摘要】 将已经序列测定的鸡传染性支气管炎病毒 ( IBV) HB株的核蛋白基因亚克隆到表达载体 pc DNA3的Kpn 酶切位点 ,快速筛选鉴定出含有 HB核蛋白基因的重组质粒 ,经酶切、PCR及 Southern鉴定为正向插入了 HB核蛋白基因 ,将其命名为 pc DNA-N。对重组质粒进行大量扩增 ,应用聚乙二醇纯化后 ,对 SPF雏鸡进行了免疫攻毒试验 ,结果表明 ,IBV核蛋白在抗感染和抗致死方面具有一定的作用。更多还原

【Abstract】 Nucleocapsid(N) Gene cDNA of isolate HB strain was amplified by RT PCR and cloned into KpnI site of plasmid pUC119. It was sequenced and subcloned into KpnI site of plasmid pcDNA3.The recombinant plasmid pcDNA N was identified by KpnI, PCR and southern hybrization.The construction of pcDNA N will provide theoretical base for IBV nucleocapsid gene immunization.更多还原
