川楝素对大鼠肾上腺嗜铬细胞胞内游离钙浓度的影响(英文)

【摘要】 在单个大鼠肾上腺嗜铬细胞上 ,采用Fura 2显微荧光测量技术 ,测量了川楝素对胞内游离钙浓度([Ca2 +]i)的影响。结果表明 ,川楝素作用于大鼠嗜铬细胞数十秒内即可导致 [Ca2 +]i 的升高。将胞外液换为无钙液或向胞外液加入钙通道阻断剂均可抑制川楝素增加 [Ca2 +]i 的作用 ,在胞外液中加入thapsigargin排空钙库后却不能阻止川楝素对 [Ca2 +]i 的作用。提示 ,川楝素可以引起细胞外Ca2 +内流 ,而对钙库似无影响。

【Abstract】 By using Fura-2 microfluorometry technique, the effects of toosendanin on the intracellular calcium concentration([Ca 2+] i) in single rat chromaffin cell were studied. The results showed that an elevation of [Ca 2+] i was found when toosendanin was puffing to the cell. Removal of Ca 2+ from the bath solution or application of nimodipine obviously inhibited the [Ca 2+] i increase induced by toosendanin, while depletion of Ca 2+ store with thapsigargin had no influence on the effect of toosendanin on [Ca 2+] i. It illustrated that toosendanin could induce the influx of extracellular calcium, but it seems that there is no effect on the calcium store.
