日本对虾血清凝集素的基本物理化学性质

【摘要】 日本对虾 (Penaeusjaponicus )亲虾体长 19~ 2 4 .5cm ,取其血清。 7种动物血液取自大白鼠、小白鼠、豚鼠、兔、鸡、羊、人。结果表明 :①虾血清分别在 2 8、37、5 6℃时热处理 30min ,其凝集活性不变 ,在 70℃和 80℃热处理 30min后凝集活性完全丧失。②血清在 pH为 5~ 7.5的TBS缓冲液中凝集活性稳定 ,在pH为 8~ 9之间凝集活性迅速下降。③凝集抑制实验表明半乳糖、甘露糖、L 阿拉伯糖、D 葡萄糖胺、N 乙酰 D 葡萄糖胺等 5种糖对血清凝集素都有较强的抑制作用 ,尤其D 葡萄糖胺和N 乙酰 D 葡萄糖胺的抑制作用更强。④交叉吸附实验结果显示日本对虾血清对大白鼠、小白鼠的红细胞有较强的亲和性

【Abstract】 The shrimp serum was collected from Penaeus japonicus, body length 19.0~24.5 cm. Seven blood samples were collected from rat, mouse, guinea pig, rabbit, chicken, sheep and human, respectively. The results show that: 1) when heated at 28, 37, and 56℃ for 30 min, respectively, the shrimp serum is stable in agglutinating activity, but when heated at 70℃ and 80℃ for 30 min, the serum losts its agglutinating activity completely; 2) at pH 5.0~7.5 of the TBS buffer, the agglutinating activity is stable, and at pH 8~9, the agglutinating activity declines dramatically; 3) the agglutinating inhibition test shows that the serum agglutination can be inhibited obviously by mannose, galactose, glucosamine(GlcN), N acetylglucosamine(GlcNAc) and arabinose, especially GlcN and GlcNAc which have the most obvious effect; 4) the crossed absorption test with the 7 animal bloods shows that the serum of P.japonicus has a strong affinity to the erythrocytes of rat and mouse.
