应激与衰老的实验研究
The Effect of Chronic Stress on Aging Process
【摘要】 目的 研究慢性应激对老化过程的影响。方法 采用悬尾方式每天予以小鼠应激刺激,持续14d,每天适当延长悬吊时间(记下小鼠体温下降情况决定所延长时间,水温维持于23±2℃)。悬吊时,每天对小鼠进行性功能监测,记录10min内跨骑、交配次数。最后一次悬尾后12h,处死小鼠,取其全脑。检测脑组织老化相关酶-单胺氧化酶(MAO-B),超氧化物歧化酶(SOD)活性,及脂褐素沉积情况。结果 悬尾6d时,小鼠跨骑与交配次数与对照组相比有显著差别(P<0.05),小鼠性功能明显衰退。悬尾12d后,脑组织MAO-B活性显著升高(P<0.05),SOD活性显著下降(P<0.01),脂褐素沉积增加(P<0.05)。结论 慢性应激易化老化进程,使小鼠呈现类老化变化。
【Abstract】 Objectives To study the effect of chronic stress on aging process.Method Hanging stress was employed to exert on 615 mice till the body temperature decreased to 30\\\\\\ in every afternoon for 14d (the water temperature remain at 23++++++ 2\\\\\\), and increased hanging tune every day according to the decrease of mice body temperature. Sexual behaviors (mounting, mating) were assessed every day in the morning.After the final stress application, the mice were sacrificed and the concentration of superoxide dismutase (SOD), monoamine oxidase (MAO- B), and the deposition level of lipofuscin was detected in mice brain using spectrophotoroetry and fiuorescentphotometry method. Result The mounting and mating numbers of stressed mice were significantly decrease at hanging stress of d6 ( P < 0.05), and the activity of SOD in brain was significantly decreased ( P < 0.01), the activity of aging - related enzyme MAO - B was enhanced (P<0.05) .Besides, deposition of lipofuscin was significantly increased (P<0.05) . Conclusion These data showed that chronic stress declined the sexual behaviour, and decrease the function of anti -oxidative impairment, The result showed that chronic stress could accelerate the aging process.
